GlyNAC’s small trial cannot establish reversal of aging
A 16-week study randomized 24 older adults—12 per arm—and reported many biomarker and function measures. Its size, multiplicity and exploratory analyses do not prove that GlyNAC reverses aging or extends healthspan.

What researchers did
Twenty-four adults age 61 to 80 with BMI above 27 kg/m² were randomized to glycine 100 mg/kg/day plus N-acetylcysteine 100 mg/kg/day or isonitrogenous alanine. Twelve younger adults formed a nonrandomized reference group and received GlyNAC for two weeks.
What they found
The publication reports changes across glutathione, oxidative stress, mitochondrial measures, inflammation, insulin resistance, endothelial function, physical-function tests, body composition, blood pressure and exploratory aging hallmarks. The registry identifies muscle glutathione concentration as the primary outcome.
How strong is the evidence?
Randomization is useful, but 12 participants per arm and many measured outcomes create substantial uncertainty. Six-minute walk changed from 522.5 to 564.8 m in the active arm (P=.053) and 518.3 to 527.1 m in placebo (P=.99); within-arm changes are not a confirmed between-group effect.
What this does not show
The trial does not establish reversal of biological aging, disease prevention, disability prevention, cognitive preservation, longer healthspan or longer life. Aging-hallmark analyses added after trial completion were explicitly exploratory.
Safety, regulation and conflicts
No supplement-related adverse events were reported and monitored liver and kidney markers were stable over 16 weeks, but 12 treated older adults cannot establish uncommon, interaction or long-term risks. FDA does not approve dietary supplements for safety or effectiveness before sale. The paper reported NIH/NIA and McNair support and declared no conflicts.
What happens next
Independent preregistered trials need larger samples, a limited set of clinically meaningful primary outcomes, correction for multiple testing, longer follow-up and direct randomized between-group estimates. A separate 114-person study lasted only two weeks and did not replicate broad function or aging claims.
